چکیده

After the oral administration of nicotinamide to human subjects, two ultraviolet-absorbing compounds have regularly been detected on chromatograms of plasma extracts. The less polar of these, named Compound Y, has been identified as N’-methyl2-pyridone-5-carboxamide (1). In the present communication, evidence is presented that the more polar metabolite, Compound X, is N’-methyl-4-pyridone-3-carboxamide.1

N-Methyl-4-pyridone-5-carboxamide as a metabolite of nicotinic acid in man and monkey.

In previous experiments reported from this laboratory (1) N-methyl-4-pyridone-5-carboxamide was isolated from rat urine and identified as the major urinary metabolite of the pyridine nucleotides. Recently Wieland et al. (2) synthesized this 4-pyridone and have found their synthetic product to be identical with the compound isolated from rat urine. Experiments have now been carried out to determ...

Purification and properties of the aldehyde oxidases from hog and rabbit livers.

The aldehyde oxidases from rabbit and hog livers have been purified to homogeneity by modifications of and additions to previously published procedures. The final preparations were judged to be homogeneous on the basis of ultracentrifugal, electrophoretic, chromatographic, spectral, and kinetic criteria. A number of physical and kinetic properties of the homogeneous enzymes have been determined...

The excretion of N-methyl-2-pyridone-5-carboxamide by man following ingestion of several known or potential precursors.

The development of a rapid method for the determination of N-methyl2-pyridone-5-carboxamide (pyridone) in urine (1) has made it practical to attempt to confirm and extend the studies of Holman and de Lange (2) and others (3-5) concerning the excretion of the pyridone following the administration of various known or possible precursors to man. In the conversion of nicotinic acid to the pyridone ...

4-Pyridone-3-carboxamide-1-β-D-ribonucleoside Triphosphate (4PyTP), a Novel NAD+ Metabolite Accumulating in Erythrocytes of Uremic Children: A Biomarker for a Toxic NAD+ Analogue in Other Tissues?

We have identified a novel nucleotide, 4-pyridone 3/5-carboxamide ribonucleoside triphosphate (4PyTP), which accumulates in human erythrocytes during renal failure. Using plasma and erythrocyte extracts obtained from children with chronic renal failure we show that the concentration of 4PyTP is increased, as well as other soluble NAD(+) metabolites (nicotinamide, N(1)-methylnicotinamide and 4Py...

Metabolic response of humans to ingestion of nicotinic acid and nicotinamide.

The identification of nicotinamide-N1-oxide as a metabolite in the urine of a schizophrenic patient prompted a study of the relative metabolism of nicotinic acid and nicotinamide in mental patients and healthy volunteers. Metabolites quantified included N1-methyl-2-pyridone-5-carboxamide, N1-methyl-4-pyridone-3-carboxamide, N1-methylnicotinamide, nicotinuric acid, and nicotinamide-N1-oxide. Mor...
